Abstract

We describe a novel route to hydrogel formation as well as polycysteine synthesis through native chemical ligation (NCL). Common thioester byproducts of NLC are cytotoxic, limiting its use in biological systems. Utilizing cysteine-thiolactone, we hope to afford an efficient and safe hydrogel via NCL by β-thiolactone ring opening of bis-cysteine terminated peptide crosslinkers. Additionally, we describe a potential method to polycysteine tail formation off functionalized polyethylene glycol (PEG) by inducing a polymerization cascade through NCL. These results describe the formation of the cysteine-thiolactone and PEG modification.
